I'd like to update my 2001 Ram 2500 5.9 360. Would this carb work with the existing fuel pump? If so, what intake would I need to make this happen?

Is this system compatible with a ford 302 engine? What other changes will be required?
Yes, as long as you have a square bore intake flange for this to bolt up to- it would fit perfectly fine.

will this system handle turbo boost
No but the super sniper EFI kit, #22772 is rated and designed for boost.

In the description states compatible with 700R4. How does the Sniper EFI system communicate with a 700R4 transmission? What additional harnesses of linkages are needed?
The 700r4 does not require a transmission computer. It does require a tv cable. Page 7 in the manual illustrates where it will connect just like it would to a carburetor

I'm building a engine and my ? Is there a lot of adjust and ect you need to due with this when engine finally get up to operating temp. My ? Is can I break in my new flat tappet came at first start up with this system and then do the adjustments
