If I use this as a primer/sealer immediately before topcoat, how fine must the substrate be sanded?

I would sand with 400 grit if applying solid paints and 600 if applying metallic.

Can this epoxy primer be used as a sealer for wood projects

This Epoxy Primer is a DTM (direct to metal) primer and is designed to be sprayed on steel, aluminum and fiber glass. Unfortunately we have no information on its performance on wood.

I saw it says 140 sq ft is that 1 coat or 2 coats?

The Eastwood Epoxy Primer(511227) is a 1:1 mix product. one Gallon of Epoxy Primer to One Gallon of Epoxy Primer Catalyst(50245ZP) combined they make 2 gallons of sprayable primer with a coverage of 280 total square feet. Spraying one medium wet coat you will get approximately 140 square feet per gallon not thinned/reduced. You can reduce this primer with a good universal urethane reducer such as Eastwood's Zero Voc Urethane Reducer(50104ZP) should you desire to reduce film build or improve flow out.

1 gal of epoxy to 1 gal of catalyst for a 1:1 mix = 2 gals sprayable. The approx. coverage listed is 140sq ft. per gallon. Is that approx coverage per sprayable gallon? I need to spray about 500 sq ft.
